Introduction
This Cryoprotectant Storage Buffer is engineered to prevent ice crystallization, maintaining tissue flexibility and integrity even at sub-zero temperatures. By inhibiting structural damage at the cellular level, it effectively preserves the antigenicity of the samples. The process is fully reversible: once retrieved, the buffer can be rinsed away, allowing the specimens to be seamlessly re-processed for either OCT cryo-embedding or paraffin infiltration.
Protocol
Tissue Storage Procedure:
-Immerse:
Following complete fixation, submerge the tissue specimens in the solution and incubate at 4°C overnight.
-Cryopreservation:
Once the tissue is fully equilibrated (thoroughly infiltrated), transfer the samples to -20°C for long-term storage.
Specimen Recovery & Downstream Processing
-Buffer Exchange:
Upon retrieval, wash the specimens four times for 5 minutes each using pre-chilled PBS to ensure complete removal of the storage buffer.
(Note: Use a PBS volume at least 10 times the volume of the tissue).
-Re-processing:
For Frozen Sectioning: Proceed with sucrose dehydration (cryoprotection) followed by OCT embedding.
For Formalin-Fixed Paraffin-Embedded (FFPE) Processing: Initiate the standard paraffin embedding workflow.
